384-well plate format using NanoBRET Ternary Complex Formation Assay

Assay Protocol

HEK293 cells transiently co-expressing NanoLuc-BRD4 and HaloTag-CRBN or HaloTag-VHL Fusion Vectors were seeded into the wells of 384-well plates. The cells were treated with reference PROTAC dBET6 or MZ1 for 4 hours. The BRET signal was measured on an Envision 2104 Multilabel Reader. BRET ratio was calculated and plotted using the GraphPad Prism program.
