HEK-293 cells co-expressing p53-HaloTag and MDM2-NanoLuc vectors were plated in 384-well plate and pre-treated with/without HaloTag NanoBRET 618 Ligand. The cells were then treated with various concentrations of reference compound Nutlin-3 for 22 hours before addition of Nano-Glo Luciferase Assay Substrate and NanoBRET measurement on an Envision 2104 Multilabel Reader. IC50 curve was plotted and IC50 value was calculated using the GraphPad Prism program.
