Human AR Reporter Cell Line

- principle: The AR Reporter Cells include the luciferase reporter gene functionally linked to a bona fide AR–responsive promoter. Thus, quantifying changes in luciferase expression in the treated reporter cells provides a sensitive surrogate measure of the changes in AR activity (INDIGO biosciences).

Human AR Reporter cells were plated in a 96-well plate and then stimulated with various concentrations of reference agonist 6a-FIT-R for 24 hours. The luminescence signal was measured with an EnVision 2104 Multilabel Reader. AR activation dose-response curve was plotted using the GraphPad Prism program.
Human AR Reporter cells were plated in a 96-well plate and then stimulated with various concentrations of reference antagonist Enzalutamide and a fixed concentration of agonist 6a-FIT-R for 24 hours. The luminescence signal was measured with an EnVision 2104 Multilabel Reader. AR activation dose-response curve was plotted using the GraphPad Prism program.
